چکیده

Materials and Methods Transgenic mice. Transgenic mice homozygous for a human PrP 129V transgene array and murine PrP null alleles (Prnp) designated Tg(HuPrP129V Prnp)-152 mice (129VV Tg152 mice) and transgenic mice homozygous for a human PrP 129M transgene array and murine PrP null alleles (Prnp) designated Tg(HuPrP129M Prnp)-35 mice (129MM Tg35 mice) have been described previously (S1-S4). Inbred FVB/NHsd mice were supplied by Harlan UK, Ltd.

Mutant Prion Protein Expression Causes Motor and Memory Deficits and Abnormal Sleep Patterns in a Transgenic Mouse Model

A familial form of Creutzfeldt-Jakob disease (CJD) is linked to the D178N/V129 prion protein (PrP) mutation. Tg(CJD) mice expressing the mouse homolog of this mutant PrP synthesize a misfolded form of the mutant protein, which is aggregated and protease resistant. These mice develop clinical and pathological features reminiscent of CJD, including motor dysfunction, memory impairment, cerebral P...

Prion diseases of humans and animals: their causes and molecular basis.

Prion diseases are transmissible neurodegenerative conditions that include Creutzfeldt-Jakob disease (CJD) in humans and bovine spongiform encephalopathy (BSE) and scrapie in animals. Prions appear to be composed principally or entirely of abnormal isoforms of a host-encoded glycoprotein, prion protein. Prion propagation involves recruitment of host cellular prion protein, composed primarily of...

INVENTORY OF NATIONAL RESEARCH ACTIVITIES IN TRANSMISSIBLE SPONGIFORM ENCEPHALOPATHIES (TSEs) IN EUROPE

Sporadic Creutzfeldt-Jakob disease (CJD) is the commonest human spongiform encephalopathy. The CJD Surveillance Project in the UK has recently extended the phenotype of this disorder by the identification of (1) a new variant of CJD in young patients, and (2) a subset of older sporadic CJD patients with accumulation of protease-resistant prion protein (PrPRES) in the brainstem and spinal cord. ...

The many faces of human prion diseases in Belgium and the world.

Prion diseases are rare neurodegenerative disorders that always lead to death and that can be transmissible under certain conditions. Although sporadic Creutzfeldt-Jakob's disease (CJD) is the best known human variant of these transmissible spongiform encephalopathies with an incidence of about 1 in 106 inhabitants, several other types of human prion disease have been described (e.g. Familial C...

Dissociation of pathological and molecular phenotype of variant Creutzfeldt-Jakob disease in transgenic human prion protein 129 heterozygous mice.

All neuropathologically confirmed cases of variant Creutzfeldt-Jakob disease (vCJD), characterized by abundant florid plaques and type 4 disease-related prion protein (PrP(Sc)) in the brain, have been homozygous for methionine at polymorphic residue 129 of PRNP. The distinctive neuropathological and molecular phenotype of vCJD can be faithfully recapitulated in Prnp-null transgenic mice homozyg...
